Test Format: DAS ELISA
Reactivity: This ELISA detects all strains known as apple mosaic or rose mosaic tested. Reaction of the ELISA is moderately strong. Optical Density at 405nm is in arrange of 0.600 - 1.400 depending on the virus titer in the samples tested. For obtaining good test result, special attention should be given to the grinding and extraction of the sample.
Sensitivity: Sensitivity of the ELISA is relatively high. The virus can be consistently detected in infected plant tissues diluted at 1:810.
Specificity: This test system has no cross
reaction with healthy plant tissues such as
apple and rose. Background is low on all of the negative control wells.

Virus Infection: Apple mosaic virus (ApMV) is a small, isometric, RNA virus which is mainly spread by grafting propagation of host plants. The virus is easily transmitted by mechanical inoculation among herbaceous hosts although it is difficult to transmit from woody to herbaceous plants.
ApMV causes a variety of leaf symptoms and reductions in fruit yield of varying severity in apple. Typical symptoms of ApMV infection in rose is leaf mosaic. Other symptoms include line patterns, ringspots, and mottle in infected rose leaves. A good indexing program is the most effective method to control the viral infection.
